The Paris Cardiomyopathy Center, coordinated by Prof. Karim Wahbi, Prof. Albert Hagège, and Prof. Damien Bonnet, conducts clinical and translational research dedicated to genetic cardiomyopathies, with the aim of developing precision medicine strategies tailored to patients.

Our team runs three rare disease reference centers within the AP-HP:

  • Hôpital Européen Georges Pompidou (HEGP): hypertrophic cardiomyopathies,
  • Hôpital Cochin: dilated cardiomyopathies, neuromuscular and metabolic cardiomyopathies,
  • Hôpital Necker Enfants Malades: pediatric and neonatal cardiomyopathies.

These centers monitor more than 2,000 patients per year, with complementary expertise by age and theme, ensuring a smooth transition from childhood to adulthood.

Structural projects 
Systematic genetic sequencing, combined with the inclusion of patients in national and international registries backed by DNA banks and biobanks (among those coordinated by our team, REMY/GEREMY for hypertrophic cardiomyopathy (HCM), OPALE or DMD-Heart-Registry for dilated cardiomyopathies (DCM) linked to LMNA variants and DMD), studies of genotype-phenotype correlations, and the identification of prognostic factors for arrhythmias and heart failure, as well as relevant biomarkers for clinical trials.

  • European LMNA-CMD Registry: improving cardiac risk stratification and developing new prognostic biomarkers in LMNA-related dilated cardiomyopathies.
  • Development of biomarkers for innovative therapies in Duchenne muscular dystrophy (DMD)
  • Genotype-phenotype studies, risk stratification, and identification of modifier genes in pediatric and adult hypertrophic cardiomyopathy 
  • Assessment of myocardial stiffness in hypertrophic cardiomyopathy

Expertise

  • Cohort analysis using cardiac MRI, advanced echocardiography, and biobanks (plasma, serum, DNA, cardiac tissue from transplants or assist devices).
  • Translational research on animal models (mice, rats, dogs) to evaluate gene therapies such as microdystrophin for DMD or MAPK inhibitors for LMNA.
  • International clinical trials: myosin inhibitors (mavacamten/aficamten), AAV9 gene therapy (TN-201), prophylactic nebivolol.
  • Integration of innovative technologies such as:
    • 18F-FAPI nuclear imaging to detect active myocardial fibrosis,
    • Deep proteomics using Imagine's Proteograph platform,
    • Shear wave imaging for routine measurement of myocardial stiffness,
    • OMICs analyses to identify modifier genes and predictive biomarkers.
